High mountain grasslands offer multiple goods and services to society but are severely threatened by improper land use practices such as abandonment or rapid intensification. In order to reduce abandonment and strengthen the common extensive agricultural practice a sustainable land use management of high mountain grasslands is needed. A spatially detailed yield assessment helps to identify possible meadows or, on the contrary, areas with a low carrying capacity in a region, making it easier to manage these sites. Such assessments are rarely available for remote and inaccessible areas. Remotely sensed vegetation indices are able to provide valuable information on grassland properties. These indices tend, however, to saturate for high biomass. This affects their applicability to assessments of high-yield grasslands.The main aim of this study was to model a spatially explicit grassland yield map and to test whether saturation issues can be tackled by consideration of plant species composition in the modelling process. The high mountain grassland of the subalpine belt (1800 – 2500 m a.s.l.) in the Kazbegi region, Greater Caucasus, Georgia, was chosen as test site for its strong species composition and yield gradients.We first modelled the species composition of the grassland described as metrically scaled gradients in the form of ordination axes by random forest regression. We then derived vegetation indices from Rapid Eye imagery, and topographic variables from a digital elevation model, which we used together with the multispectral bands as predictive variables. For comparison, we performed two yield models, one excluding the species composition maps and one including the species composition map as predictors. Moreover, we performed a third individual model, with species composition as predictors and a split dataset, to produce the final yield map.Three main grassland types were found in the vegetation analysis: Hordeum violaceum-meadows, Gentianella caucasea-grassland and Astragalus captiosus-grassland. The three random forest regression models for the ordination axes explained 64%, 33% and 46% of the variance in species composition. Independent validation of modelled ordination scores against a validation data set resulted in an R2 of 0.64, 0.32 and 0.46 for the first, second and third axes, respectively. The model based on species composition resulted in a R2 = 0.55, whereas the benchmark model showed weaker relationships between yield and the multispectral reflectance, vegetation indices, and topographical parameters (R2 = 0.42). The final random forest yield model used to derive the yield map resulted in 62% variance explained and an R2 = 0.64 between predicted and observed biomass. The results further indicate that high yields are generally difficult to predict with both models.The benefit of including a species composition map as a predictor variable for grassland yield lies in the preservation of ecologically meaningful features, especially the occurrence of high yielding vegetation type of Hordeum violaceum meadows is depicted accurately in the map. Even though we used a gradient based design, sharp boundaries or immediate changes in productivity were visible, especially in small structures such as arable fields or roads (Fig. 6b), making it a valuable tool for sustainable land use management. The saturation effect however, was mitigated by using species composition as predictor variables but is still present at high yields.  相似文献

In recent years, the assessment of ecosystem services (ES) supply has been based on the use of Land Use/Land Cover (LULC) data as proxies for spatial representation of ecosystems. Nevertheless, some shortcomings of this method, such as uncertainties derived from generalization of the ecosystem types and assumptions of invariance across spatial scales, indicate the need for new approaches. Such approaches could be aimed at improving knowledge of the relationships between ecosystem services and landscape structure and the spatial characteristics of ES patterns. In this study, we propose an integrative approach that involves the generation and analysis of continuous maps representing the supply of five ES potentially related to the amount of biomass. Five remote sensing images of the Northwestern Iberian Peninsula, obtained with Landsat-5 TM, were used to generate a proxy for net primary production by combining the normalized difference vegetation index (NDVI) of each image to calculate a ΣNDVI index that could act as a potential indicator of some ecosystem services. This information was combined with three variables – terrain slope, population density and occurrence of protected areas – to produce spatial models for the five ES and eventually a series of five supply maps. Food, materials and energy provision services showed a clustered pattern, with high supply values in flat zones and areas with high population densities. In contrast, mass flow and climate regulation services were more widely distributed throughout the study area. The five ecosystem service patterns were analyzed at different scales by two methods: lacunarity and four term local quadrat variance (4TLQV) analysis. These methods revealed differences in the spatial pattern: lacunarity analysis was useful for detection of scale thresholds at the local level, whereas 4TLQV was more sensitive to scale thresholds at larger spatial levels. Thus, the variance analysis yielded higher values for larger windows sizes, particularly for provisioning services. The results demonstrated the suitability of the proposed approach for the spatially explicit modeling of ecosystem services, avoiding the uncertainty of other assessments such as those based on LULC data, and for the exploratory analysis of ES supply from a spatial point of view.  相似文献

1. Digital video recording (DVR) devices, such as the GoPro Hero, have the potential to greatly benefit pollination ecology, but the advantages of digitally recording pollinator activity over direct human observation have not been formally assessed. 2. Two plant taxa, Lavandula angustifolia and Canna ‘sp.’, with differing floral morphology, were used to compare the value of DVR and direct observations in estimating honeybee (Apis mellifera) visitation, flower density and number of flowers visited per foraging bout. 3. The two methods yielded identical results when observing the structurally simple L. angustifolia at both high (10.54 ± 0.52 per plant) and low honeybee density (2.24 ± 0.20 per plant). However, DVR underestimated the number of flowers scored in the field of view (28.7 ± 1.8 direct vs. 22.7 ± 0.9 DVR), the number of honeybees observed (5.3 ± 0.8 direct vs. 3.7 ± 0.7 DVR) and the number of flowers visited during foraging bouts (8.3 ± 1.2 direct vs. 5.5 ± 1.0 DVR) on the more complex Canna ‘sp.’ 4. It is concluded that portable weatherproof DVR devices such as the GoPro Hero are valuable tools for pollination biologists, allowing a single researcher to make simultaneous observations of multiple plants in one or more sites, whilst also allowing the footage to be reviewed. However, DVR devices are limited by their depth and field of view when target plants are large or structurally complex.  相似文献

An objective, quantifiable index of structural biodiversity that could be rapidly obtained with reduced or no field effort is essential for the use of structure as universal ecological indicator for ecosystem management. Active remote sensing provides a rapid assessment tool to potentially guide land managers in highly dynamic and spatially complex landscapes. These landscapes are often dependent on frequent disturbance regimes and characterized by high endemism.We propose a modified Shannon–Wiener Index and modified Evenness Index as stand structural complexity indices for surrogates of ecosystem health. These structural indices are validated at Tall Timbers Research Station the site of one of the longest running fire ecology studies in southeastern U.S. This site is dominated by highly dynamic pine-grassland woodlands maintained with frequent fire. Once the dominant ecosystem in the Southeast, this woodland complex has been cleared for agriculture or converted to other cover types, and depends on a frequent (1- to 3-year fire return interval) low- to moderate-intensity fire regime to prevent succession to mixed hardwood forests and maintain understory species diversity. Structural evaluation of the impact of multiple disturbance regimes included height profiles and derived metrics for five different fire interval treatments; 1-year, 2-year, 3-year, mixed fire frequency (a combination of 2- and 4-year fire returns), and fire exclusion. The 3-dimensional spatial arrangement of structural elements was used to assess hardwood encroachment and changes in structural complexity. In agreement with other research, 3-year fire return interval was considered to be the best fire interval treatment for maintaining the pine-grassland woodlands, because canopy cover and vertical diversity indices were shown to be statistically higher in fire excluded and less frequently burned plots than in 1- and 2-year fire interval treatments. We developed a LiDAR-derived structural diversity index, LHDI, and propose that an ecosystem-specific threshold target for management intervention can be developed, based on significant shifts in structure and composition using this new index.Structural diversity indices can be valuable surrogates of ecosystem biodiversity, and ecosystem-specific target values can be developed as objective quantifiable goals for conservation and ecosystem integrity, particularly in remote areas.  相似文献

Pseudomonas sp. StFLB209 was isolated from potato leaf as an N-acylhomoserine lactone (AHL)-producing bacterium and showed a close phylogenetic relationship with P. cichorii, a known plant pathogen. Although there are no reports of potato disease caused by pseudomonads in Japan, StFLB209 was pathogenic to potato leaf. In this study, we reveal the complete genome sequence of StFLB209, and show that the strain possesses a ppuI-rsaL-ppuR quorum-sensing system, the sequence of which shares a high similarity with that of Pseudomonas putida. Disruption of ppuI results in a loss of AHL production as well as remarkable reduction in motility. StFLB209 possesses strong pectate lyase activity and causes maceration on potato tuber and leaf, which was slightly reduced in the ppuI mutant. These results suggest that the quorum-sensing system is well conserved between StFLB209 and P. putida and that the system is essential for motility, full pectate lyase activity, and virulence in StFLB209.  相似文献

The Cecropia genus is widely distributed in Latin America including at least 60 species, and some of them are commonly used in traditional medicine for the treatment of several diseases. We used Cecropia pachystachya Trécul to search for quorum sensing (QS) inhibitors compounds and found that the aqueous extract of C. pachystachya leaves is a promising source of substances with this activity. Using as biosensor Chromobacterium violaceum ATCC 31532 and Escherichia coli pSB403, the compounds chlorogenic acid (2), isoorientin (3), orientin (4), isovitexin (6), vitexin (7), and rutin (9) were identified as QS inhibitors. None of these compounds inhibited the growth of neither the used biosensors nor the microorganisms Staphylococcus aureus ATCC 23591, Escherichia coli ATCC 25922 and Saccharomyces cerevisiae, used here as growth inhibition controls. Along with the rutin, here we presented for the first time the QS-inhibition potential of the C-glycosyl flavonoids. The prospective of this evidence lead to the use of these compounds as antipathogenic drugs or antifoulants.  相似文献

Measurements on modern coral reefs at Lee Stocking Island (Bahamas) illustrate that boring cyanobacteria species make a major contribution to microboring bioerosion rates. Borings attributed to cyanobacteria also occur in fossil environments. Bioerosional studies on Permian and Triassic reefs show similar intensities to those observed on modern equivalents. The importance of borings assigned to cyanobacterial activity is even more apparent in paleobathymetry. Comparison of the bathymétrie ranges known from modern and fossil microborings demonstrates a preference of boring cyanobacteria for shallow marine environments. Furthermore, some traces are linked to distinct portions of the shallow euphotic zone. They significantly contribute to characterize typical microboring assemblages, which are used for paleodepth reconstructions. In contrast to these stenobathic species, one cyanobacterial species turned out to be eurybathic. It has been recorded as deep as the dysphotic zone but may even extend to the aphotic zone.  相似文献

Luminescence Resonance Energy Transfer, or LRET, is a powerful technique used to measure distances between two sites in proteins within the distance range of 10-100 Å. By measuring the distances under various ligated conditions, conformational changes of the protein can be easily assessed. With LRET, a lanthanide, most often chelated terbium, is used as the donor fluorophore, affording advantages such as a longer donor-only emission lifetime, the flexibility to use multiple acceptor fluorophores, and the opportunity to detect sensitized acceptor emission as an easy way to measure energy transfer without the risk of also detecting donor-only signal. Here, we describe a method to use LRET on membrane proteins expressed and assayed on the surface of intact mammalian cells. We introduce a protease cleavage site between the LRET fluorophore pair. After obtaining the original LRET signal, cleavage at that site removes the specific LRET signal from the protein of interest allowing us to quantitatively subtract the background signal that remains after cleavage. This method allows for more physiologically relevant measurements to be made without the need for purification of protein.  相似文献

Abstract Genome analysis of Pseudomonas aeruginosa was performed by digestion with rare-cutting restriction endonucleases and subsequent one- and two-dimensional field inversion gel electrophoresis (FIGE). The frequency of chromosomal recognition sites increased in the order Spe I, Dra I, Xba I, Ssp I, Nhe I. The genome size of strain PAO and the 17 IATS strains varied from 4.4 × 106 to 5.4 × 106 base pairs. Double restriction digests and two-dimensional FIGE provide a genome fingerprint which is useful for the identification and typing of the respective strains.  相似文献
